The more common form (85% of patients) of hereditary angioneurotic edema is due to an absolute decrease in the amount of C 1 esterase inhibitor. A less common form (15% of patients) is due to a functional defect where quantitative levels may be normal. C1-esterase inhibitor reacts to the specific antibody forming insol-uble immune complexes. The turbidity caused by these immune complexes is proportional to C1-esterase inhibitor concentration in the sample and may be spectrophotometrically measured. PROVIDED REAGENTS A. Reagent A: phosphate buffer, pH 7.4. Active Figure (Chapter 11, p.383) A base solution is added from the buret to the acid solution in the beaker. The indicator phenolphthalein changes from colorless, its color in acidic solutions, to pink, its color in basic solutions, when the reaction reaches completion. PLACE PAGE DESCRIPTION HERE. Its main function is the inhibition of the complement system to prevent spontaneous activation. C1-inhibitor is an acute-phase protein that circulates in blood at levels of around 0.25 g/L.The levels rise ~2-fold during inflammation. Patient Preparation Collect Serum separator tube. Specimen Preparation Separate serum from cells ASAP or within 2 hours of collection. Transfer three 1 mL aliquots of serum to individual ARUP Standard Transport Tubes and freeze immediately. (Min: 0.5 mL/tube) Hereditary angioedema (HAE) is a disorder that results in recurrent attacks of severe swelling. The swelling most commonly affects the arms, legs, face, intestinal tract, and airway. Itchiness does not typically occur. If the intestinal tract is affected abdominal pain and vomiting may occur. Swelling of the airway can result in its obstruction. Manual de Chimie
